Overview

WX390 + toripalimab is a **combination therapy** consisting of WX390, an orally administered, highly potent dual inhibitor of PI3K and mTOR, and toripalimab, an intravenous anti-PD-1 monoclonal antibody. WX390 suppresses tumors by targeting pan-PI3K and mTOR kinase activities, which reduces immunosuppressive tumor microenvironment components such as regulatory T cells (Treg) and myeloid-derived suppressor cells (MDSC), thus potentially enhancing the activity of immune checkpoint inhibitors like toripalimab. Toripalimab blocks the PD-1 immune checkpoint, reinvigorating T-cell mediated antitumor responses. The combination is under clinical investigation for advanced solid tumors and has shown early evidence of efficacy and manageable safety profiles, particularly in cervical, head and neck, and urothelial cancers.
