Overview

WXD171-HlaH35L is an experimental recombinant probiotic vaccine candidate designed for the prevention of Staphylococcus aureus infections. It utilizes the probiotic bacterium Limosilactobacillus reuteri (strain WXD171) as a live delivery vehicle, genetically engineered to express HlaH35L on its surface. HlaH35L is a detoxified mutant form of the S. aureus alpha-hemolysin (Hla) toxin, where a single amino acid substitution (histidine 35 replaced with leucine) abolishes its hemolytic activity while retaining its immunogenic properties. Administered orally, the vaccine is intended to stimulate mucosal and systemic immune responses, including the production of secretory IgA and IL-17A, to protect against localized infections such as pneumonia and skin infections, as well as systemic infections like sepsis, including those caused by methicillin-resistant S. aureus (MRSA).
