Overview

WYC-209 is a synthetic retinoic acid analog (retinoid) that functions as an epigenetic modulator and a potent inhibitor of cancer stem cells (CSCs). It is primarily recognized for its ability to sensitize multiple myeloma (MM) cells to proteasome inhibitors, such as carfilzomib, by upregulating the expression of ZMYND8 (Zinc finger MYND domain-containing protein 8). This upregulation is achieved through the remodeling of the epigenetic landscape, specifically by increasing H3K27 acetylation (H3K27ac) at the ZMYND8 locus. Preclinical studies have demonstrated that WYC-209 synergistically induces apoptosis, inhibits cell viability, and exacerbates endoplasmic reticulum stress in malignant cells. Beyond multiple myeloma, WYC-209 has shown efficacy in inducing apoptosis in CSCs across various solid tumors, including breast cancer, lung cancer, and melanoma, while maintaining a favorable safety profile in non-malignant cells.
