WYE-132 is a small molecule dual inhibitor of the mammalian target of rapamycin (mTOR) complexes 1 and 2 (mTORC1/2). Developed by Wyeth (now part of Pfizer), it has been investigated primarily in preclinical models for its anti-tumor activity, particularly in epithelial ovarian cancer. WYE-132 exerts its effects by blocking the phosphorylation of downstream mTOR targets, leading to the inhibition of cell proliferation and the induction of caspase-dependent apoptosis. Beyond direct mTOR inhibition, WYE-132 has been shown to inhibit sphingosine kinase-1 (SphK1) activity, resulting in the production of pro-apoptotic ceramide. In vivo studies have demonstrated its potential to inhibit tumor growth both as a monotherapy and in combination with chemotherapy agents like paclitaxel.
