WYE-354 is a **synthetic, potent, ATP-competitive small molecule inhibitor of the mammalian target of rapamycin (mTOR) kinase**. It specifically inhibits both mTORC1 and mTORC2 complexes, leading to the downregulation of mTOR signaling and subsequent antiproliferative effects in cancer models. WYE-354 has shown the ability to reverse multidrug resistance in acute myeloid leukemia by inhibiting ABCB1-mediated drug efflux, and significantly reduces tumor growth, cell migration, and cell viability in preclinical models, including gallbladder cancer. Its selectivity for mTOR is over 100-fold greater than for PI3K[1][2][3][5].
