WZ35 is a novel small molecule curcumin derivative that functions as a glycolysis inhibitor with potential therapeutic applications in hepatocellular carcinoma (HCC) and gastric cancer. It exerts its anti-cancer effects by inducing oxidative stress and activating the ROS-dependent c-Jun N-terminal kinase (JNK) pathway, leading to endoplasmic reticulum (ER) stress and apoptosis. Mechanistically, WZ35 prevents the nuclear translocation of Yes-associated protein (YAP), which subsequently downregulates the transcription of Glucose transporter 1 (GLUT1). This disruption of the YAP-GLUT1 axis results in metabolic reprogramming characterized by impaired glycolysis, oxidative phosphorylation, and purine metabolism, effectively inhibiting tumor cell proliferation and growth in preclinical models.
