X-PEG-PAL is a PEGylated recombinant phenylalanine ammonia lyase (PAL) enzyme being developed by SunBio for the treatment of phenylketonuria (PKU). PKU is a rare metabolic disorder caused by a deficiency in the enzyme phenylalanine hydroxylase, leading to the accumulation of phenylalanine to neurotoxic levels. X-PEG-PAL serves as an enzyme replacement therapy that bypasses the deficient hydroxylase pathway by converting phenylalanine into trans-cinnamic acid and ammonia, which are then excreted. The addition of polyethylene glycol (PEG) moieties to the PAL enzyme is designed to enhance its stability, reduce immunogenic potential, and prolong its half-life in circulation, potentially offering a therapeutic option for patients who require significant reduction in blood phenylalanine levels.
