X15 is a novel, potent, and selective covalent inhibitor of Janus kinase 3 (JAK3). It is being investigated for the treatment of autoimmune and inflammatory diseases, specifically atopic dermatitis. By covalently binding to JAK3, it inhibits the signaling pathways of pro-inflammatory cytokines such as IL-2 and IFN-γ, subsequently reducing the production of IL-2 and IL-6. In preclinical mouse models of atopic dermatitis, X15 demonstrated significant efficacy in reducing skin inflammation and ear thickness. It exhibits a favorable pharmacokinetic profile, including oral bioavailability and a relatively long half-life of approximately 20 hours.
