X4-136 is an orally available small molecule inhibitor of the C-X-C chemokine receptor type 4 (CXCR4), a chemokine receptor involved in cell migration, tumor growth, metastasis, and inflammation. It is closely related to mavorixafor and is under preclinical and early clinical evaluation for its potential to enhance the efficacy of established therapies in diseases such as aggressive pediatric acute lymphoblastic leukemia (ALL), cervical cancer, and other malignancies. X4-136 functions primarily by blocking the CXCL12/CXCR4 signaling pathway, suppressing tumor progression, reducing metastasis, and modulating immune microenvironments. It has demonstrated improved outcomes when combined with chemotherapy and immunotherapies in preclinical models[1][3][4][5][8][9].
