XB002 is a next-generation antibody-drug conjugate (ADC) targeting Tissue Factor (TF), a protein frequently overexpressed in various solid tumors and associated with poor prognosis. Developed by Exelixis through a license from Iconic Therapeutics, XB002 utilizes a proprietary linker-payload technology designed to improve the therapeutic index compared to first-generation TF-targeting ADCs. The antibody component binds to TF without interfering with the coagulation cascade, minimizing potential bleeding risks. Upon binding and internalization, the cytotoxic payload—a microtubule inhibitor—is released to induce cell death. It is currently being evaluated in Phase 1/2 clinical trials (JEWEL-101) as a monotherapy and in combination with other agents like nivolumab or bevacizumab for advanced solid tumors, including non-small cell lung cancer, squamous cell carcinoma of the head and neck, and esophageal cancer.
