XB002 + nivolumab is an investigational combination therapy for advanced solid tumors. XB002 is a novel antibody-drug conjugate composed of a high-affinity tissue factor-directed human monoclonal antibody conjugated to the auristatin-based payload zovodotin using a stable protease-cleavable linker. Nivolumab is a fully human IgG4 monoclonal antibody targeting programmed cell death protein 1 (PD-1), approved as "Opdivo". The combination is being studied to enhance antitumor efficacy, with XB002 delivering cytotoxic payload directly to TF-expressing tumor cells, and nivolumab reactivating antitumor immune response. The combination is under clinical investigation (phase 1) in several advanced solid tumors, including pancreatic and esophageal cancers. XB002 is developed by Exelixis, while nivolumab (Opdivo) is developed and manufactured by Bristol Myers Squibb. Studies indicate promising safety and antitumor activity for this combination, with the potential to treat difficult-to-treat tumors with high unmet needs[1][2][3][4].
