XB010

XB010 is an investigational antibody-drug conjugate (ADC) developed for the treatment of advanced solid tumors. It consists of a monoclonal antibody targeting the oncofetal antigen 5T4, which is overexpressed in many solid tumors, conjugated via Catalent’s SMARTag site-specific bioconjugation platform to a monomethyl auristatin E (MMAE) cytotoxic payload. The ADC is designed to deliver MMAE directly to tumor cells expressing 5T4, thereby inhibiting tubulin polymerization and inducing apoptosis in cancer cells. Preclinical studies have demonstrated antitumor activity in xenograft models. XB010 entered phase 1 clinical trials in 2024 as both monotherapy and in combination with pembrolizumab for various advanced or metastatic solid tumors including non-small cell lung cancer, head and neck squamous cell carcinoma, triple negative breast cancer, esophageal squamous cell carcinoma, endometrial carcinoma, and hormone receptor positive breast cancer[1][2][4][5][6][7][9][10].
