xB3-IL-1RA is a recombinant fusion protein developed by Bioasis Technologies designed to treat neuroinflammatory conditions by delivering a therapeutic payload across the blood-brain barrier (BBB). It consists of the proprietary xB3 peptide, a 12-amino acid sequence derived from melanotransferrin, fused to an interleukin-1 receptor antagonist (IL-1RA). The xB3 component facilitates transport across the BBB via receptor-mediated transcytosis targeting the low-density lipoprotein-related protein 1 (LRP1) receptor. Once in the central nervous system, the IL-1RA component inhibits the pro-inflammatory interleukin-1 (IL-1) signaling pathway by binding to the IL-1 receptor. Preclinical studies have demonstrated efficacy in animal models of multiple sclerosis and neuropathic pain, showing reduced clinical symptoms and delayed disease onset.
