Overview

xB3-siRNA is a **preclinical central nervous system RNA delivery platform** developed by Bioasis that combines the company's xB3 blood-brain barrier shuttle peptide with a small interfering RNA payload. The xB3 peptide is a 12-amino-acid melanotransferrin-derived carrier intended to cross the blood-brain barrier through receptor-mediated transcytosis via low-density lipoprotein receptor-related protein 1, enabling intracellular delivery of siRNA into the brain. In reported preclinical work, an xB3-siRNA construct directed against *NADPH oxidase 4* achieved brain exposure after systemic administration, reduced target expression, and showed neuroprotective effects in an ischemic stroke model. The term does not denote a single approved drug product; rather, it refers to a research-stage peptide-siRNA conjugate approach that could be adapted to multiple central nervous system indications.
