XDD60 is an experimental small molecule inhibitor of Ribonucleoside-diphosphate reductase subunit M2 (RRM2), identified through integrative bioinformatic screening and structure-based virtual screening. It was developed to address advanced prostate cancer, specifically castration-resistant prostate cancer (CRPC), by targeting oncogenic drivers that operate independently of the androgen receptor (AR) signaling pathway. RRM2 is a critical enzyme for DNA synthesis and is transcriptionally regulated by the RB1/E2F1 axis; its overexpression is associated with poor clinical outcomes in prostate cancer. XDD60 has demonstrated potent anti-tumor efficacy in preclinical models, exhibiting superior growth inhibition compared to traditional AR antagonists by inducing cell cycle arrest and apoptosis.
