Overview

XELOX + tislelizumab + tyrosine kinase inhibitor is an investigational combination therapy regimen being evaluated for the treatment of advanced hepatocellular carcinoma (HCC). This regimen consists of the XELOX chemotherapy backbone (comprising oxaliplatin and capecitabine), the anti-PD-1 monoclonal antibody tislelizumab, and a small-molecule tyrosine kinase inhibitor (TKI), which in clinical trial settings is typically lenvatinib or regorafenib. The combination is designed to target multiple facets of tumor biology: the XELOX components provide cytotoxic effects and induce immunogenic cell death; tislelizumab acts as a checkpoint inhibitor to restore T-cell mediated anti-tumor immunity; and the TKI component provides anti-angiogenic and anti-proliferative effects by inhibiting receptors such as VEGFR, FGFR, and PDGFR. This multi-modal approach is currently being investigated in the HCC-SIGHT platform trial led by Fudan University, specifically targeting patients with advanced HCC who have experienced first-line treatment failure.
