Overview

XEN-D0101 is an experimental small molecule antiarrhythmic agent developed by Xention Discovery for the treatment of atrial fibrillation. It functions as a selective antagonist of the voltage-gated potassium channel Kv1.5 (encoded by the KCNA5 gene), which is primarily expressed in the human atria. By inhibiting this channel, XEN-D0101 selectively prolongs the atrial refractory period, thereby preventing premature atrial contractions and helping to maintain sinus rhythm without significantly affecting ventricular repolarization or the QTc interval. Although the compound progressed into Phase I clinical trials, where it demonstrated safety and tolerability at oral doses up to 300 mg, its development has since been discontinued.
