Overview

Xenin is a 25-amino acid gastrointestinal peptide hormone that belongs to the neurotensin family. It is produced by the proteolytic cleavage of the N-terminus of the alpha-coatomer protein (alpha-COP) and is primarily secreted by enteroendocrine K-cells in the proximal small intestine, often in response to nutrient ingestion. Xenin acts as an anorexigenic signal, reducing food intake and slowing gastric emptying through both central and peripheral mechanisms. It is frequently co-secreted with glucose-dependent insulinotropic polypeptide (GIP) and has been shown to potentiate GIP-mediated insulin secretion. Recent research highlights its role in lipid metabolism, specifically its ability to directly stimulate lipolysis in white adipose tissue by increasing the phosphorylation of hormone-sensitive lipase (HSL). Due to its effects on appetite and fat metabolism, xenin and its stable analogs are being investigated as potential therapeutic agents for obesity and type 2 diabetes.
