The **xenogeneic murine tyrosinase DNA vaccine** is a plasmid DNA vaccine encoding the murine (mouse) tyrosinase gene, a melanosomal differentiation antigen involved in the biosynthesis of melanin. This vaccine is classified as a xenogeneic vaccine because it introduces a non-self (murine) version of tyrosinase to break immune tolerance and stimulate T cell and antibody responses against self (syngeneic) tyrosinase expressed on melanoma cells. It has been evaluated both in veterinary medicine for canine malignant melanoma and in human clinical trials for patients with advanced melanoma. Mechanistically, the vaccine induces **CD8+ T cell responses** and antibody responses targeting tyrosinase-expressing tumor cells, thus prompting tumor-specific cytotoxicity. The vaccine has been administered by intramuscular injection followed by in vivo electroporation to facilitate gene uptake. Trials show it is generally well-tolerated, with mainly mild injection site reactions. The platform was developed with the intent to overcome immune ignorance/tolerance to tumor antigens for improved cancer immunotherapy[1][2][4].
