Xentuzumab is a humanized IgG1 monoclonal antibody that targets and neutralizes insulin-like growth factor 1 (IGF-1) and insulin-like growth factor 2 (IGF-2) ligands. By binding to these ligands, xentuzumab inhibits their proliferative activity and blocks IGF-mediated signaling pathways involved in tumor cell growth and survival. This mechanism restores inhibition of the AKT pathway when used in combination with other agents such as everolimus. Xentuzumab has been investigated primarily for the treatment of advanced solid tumors, including hormone receptor-positive/HER2-negative breast cancer, non-small cell lung cancer (NSCLC), and metastatic castration-resistant prostate cancer[2][3][4][5][9][10].
