Overview

**Xevinapant + cisplatin** is a combination regimen under investigation primarily for the treatment of locally advanced squamous cell carcinoma of the head and neck (LA-SCCHN). Xevinapant (Debio 1143) is a first-in-class, oral small molecule antagonist of inhibitor of apoptosis proteins (IAPs), which restores cancer cell sensitivity to apoptosis. Combined with high-dose cisplatin, a platinum-based chemotherapy with DNA-crosslinking (cytotoxic) effects, and standard radiotherapy (CRT), this regimen aims to enhance tumor response by promoting programmed cell death and antitumor immunity. Clinical trials have shown improved survival and locoregional control compared with standard cisplatin-based chemo-radiotherapy, with predictable and manageable safety. Xevinapant is developed by Debiopharm, and the combination strategy is under late-phase clinical trial evaluation for head and neck cancers[1][2][4][6].
