Overview

XG004 is an investigational **topical gel** containing a single small-molecule naproxen-pregabalin prodrug conjugate developed by Xgene Pharmaceutical Group for painful osteoarthritis of the knee. Following dermal absorption and enzymatic cleavage, it is designed to deliver naproxen-mediated cyclooxygenase inhibition and pregabalin-mediated modulation of voltage-dependent calcium-channel alpha-2/delta subunits, addressing inflammatory nociceptive pain and neuropathic pain components. In a completed randomized placebo-controlled Phase 1b/2a knee-osteoarthritis study, XG004 topical gel showed acceptable tolerability and exploratory improvements in pain, physical function, and sleep-related outcomes.
