Overview

Xianglian Anchang Decoction (XLAC) is a polyherbal Traditional Chinese Medicine (TCM) formulation composed of 15 medicinal components, including *Paeonia lactiflora*, *Angelica dahurica*, *Plantago asiatica*, *Saposhnikovia divaricata*, *Poria cocos*, *Pueraria lobata*, *Aspergillus oryzae*, *Lonicera japonica*, *Aucklandia lappa*, *Panax notoginseng*, *Agrimonia pilosa*, *Cynanchum paniculatum*, *Corydalis yanhusuo*, *Scutellaria baicalensis*, and *Coptis chinensis*. It is derived from the classical Ming Dynasty formula Xianglian Wan and is primarily indicated for the treatment of ulcerative colitis (UC). XLAC exerts its therapeutic effects by targeting Toll-like receptor 4 (TLR4), which subsequently inhibits the activation of the NLRP3 inflammasome and Gasdermin D (GSDMD)-mediated pyroptosis. Additionally, the decoction promotes the restoration of intestinal barrier integrity by upregulating tight junction proteins such as ZO-1 and Occludin and normalizing goblet cell architecture and mucus secretion.
