Xilio PSMA-targeted masked T cell engager is a preclinical biologic being developed by Xilio Therapeutics for the treatment of prostate cancer. Utilizing Xilio's proprietary Advanced Tumor-Activated Cell Engager (ATACR) platform, the drug is designed as a bispecific molecule that simultaneously binds to prostate-specific membrane antigen (PSMA) on tumor cells and the CD3 component of the T cell receptor complex. A key feature of the ATACR format is the inclusion of a mask on the CD3-targeting domain, which prevents systemic T cell activation and associated toxicities, such as cytokine release syndrome. This mask is engineered to be cleaved by proteases specifically found within the tumor microenvironment, thereby restricting potent T cell-mediated cytotoxicity to the tumor site and improving the therapeutic index. Xilio nominated a development candidate for this program in the third quarter of 2025 and plans to submit an Investigational New Drug (IND) application in the first quarter of 2027.
