Xilio STEAP1-masked T cell engager

Xilio Therapeutics is developing a preclinical masked T cell engager (TCE) targeting Six-transmembrane epithelial antigen of prostate 1 (STEAP1). Utilizing Xilio's proprietary SEECR (selective effector-enhanced cell engager) platform, the molecule is a tri-specific biologic designed to redirect T cells to destroy STEAP1-expressing cancer cells. The design incorporates a masking technology that restricts T cell activation to the tumor microenvironment, aiming to improve the therapeutic index by reducing systemic side effects. Additionally, the molecule features built-in co-stimulatory signaling to enhance the potency and persistence of T cell activity and conditional half-life modulation. The program is currently in IND-enabling studies, with potential applications in prostate, colorectal, and lung cancers.
