Overview

This is a combination regimen consisting of Xilonix, Onivyde (irinotecan liposome injection), and 5-fluorouracil (5-FU), sometimes with leucovorin. **Xilonix** is a monoclonal antibody that acts as an interleukin-1 alpha antagonist, targeting inflammatory pathways involved in cancer progression. **Onivyde** is a nanoliposomal formulation of irinotecan, a topoisomerase I inhibitor that interferes with DNA replication in cancer cells. **5-Fluorouracil** is an antimetabolite chemotherapeutic agent that inhibits thymidylate synthase, disrupting DNA synthesis and cell division. This combination has been studied for the treatment of advanced or metastatic pancreatic adenocarcinoma, particularly after progression following gemcitabine-based therapy[2][5]. The regimen aims to leverage complementary mechanisms—immune modulation by Xilonix and cytotoxic effects from Onivyde and 5-FU—to improve outcomes in difficult-to-treat cancers.
