Overview

XIN5494 is a preclinical small molecule inhibitor of MAPK-activated protein kinase 2 (MK2), originally developed by XinThera and subsequently acquired by Gilead Sciences. MK2 is a downstream effector of the p38 MAPK pathway and plays a critical role in the post-transcriptional regulation of pro-inflammatory cytokines, including TNF-alpha, IL-1 beta, and IL-6. By targeting MK2, XIN5494 is designed to provide a potent anti-inflammatory effect while potentially avoiding the side effects associated with direct p38 MAPK inhibition. The drug is being investigated for the treatment of various inflammatory and autoimmune conditions, such as rheumatoid arthritis.
