Overview

XL101 is a preclinical-stage PASylated recombinant human leptin being developed by XL-protein for the treatment of metabolic diseases, including obesity and lipodystrophy. The drug utilizes XL-protein's proprietary PASylation technology, which involves the genetic fusion of the leptin protein with a conformationally disordered polypeptide sequence consisting of Proline, Alanine, and Serine (PAS). This modification increases the hydrodynamic volume of the molecule, thereby reducing renal clearance and significantly extending its plasma half-life compared to native leptin. By acting as an agonist at the leptin receptor (LEPR), XL101 aims to restore or enhance leptin signaling to regulate energy homeostasis, suppress appetite, and manage body weight. The PASylation approach is intended to provide a more stable and long-acting therapeutic profile, potentially allowing for less frequent dosing.
