XL114 is a novel small molecule anti-cancer compound that inhibits the CARD11-BCL10-MALT1 (CBM) complex, a key component of signaling downstream of B- and T-cell receptors. This pathway promotes the survival and proliferation of B- and T-cell lymphomas. By inhibiting this complex, XL114 disrupts aberrant CBM signaling implicated in hematologic malignancies such as non-Hodgkin’s lymphoma (NHL), including subtypes like activated B-cell-like diffuse large B-cell lymphoma (DLBCL), mantle cell lymphoma, chronic lymphocytic leukemia/small lymphocytic lymphoma, and potentially other blood cancers. Preclinical studies have shown activity in models resistant to Bruton's tyrosine kinase inhibitors. The drug is being developed by Exelixis following an in-license from Aurigene Discovery Technologies[1][2][5][6][8].
