Overview

XL120 is a preclinical-stage PASylated insulin being developed by XL-protein for the treatment of diabetes. The drug utilizes XL-protein's proprietary PASylation technology, which involves the genetic fusion of insulin with a conformationally disordered polypeptide sequence composed of Proline, Alanine, and Serine (PAS). This modification is designed to significantly extend the plasma half-life of the insulin molecule by increasing its hydrodynamic volume, thereby reducing renal clearance. As a long-acting insulin replacement therapy, XL120 aims to provide sustained glycemic control with a reduced frequency of administration compared to standard insulin formulations. The PAS sequence is biodegradable, offering a potential advantage over synthetic polymers like PEG.
