XL130 is a long-acting, PASylated recombinant interleukin-1 receptor antagonist (IL-1Ra) being developed by XL-protein for the treatment of inflammatory diseases, such as rheumatoid arthritis. Utilizing XL-protein's proprietary PASylation technology, the drug consists of the IL-1Ra protein genetically fused to a conformationally disordered polypeptide sequence comprising Proline, Alanine, and Serine (PAS). This modification increases the hydrodynamic volume of the molecule, significantly extending its plasma half-life compared to non-modified IL-1Ra (e.g., anakinra) by reducing renal filtration. XL130 acts as a competitive antagonist at the interleukin-1 receptor type I (IL-1R1), blocking the pro-inflammatory signaling of IL-1α and IL-1β. It is currently in preclinical development.
