Overview

XL1350 (also known as EX-1350) is an orally bioavailable, small-molecule agonist of the Stimulator of Interferon Genes (STING) protein, currently under development by Exelixis for the treatment of advanced solid tumors. STING is a key component of the innate immune system that, upon activation, induces the expression of Type I interferons and other pro-inflammatory cytokines. By targeting STING, XL1350 aims to transform the tumor microenvironment from immunologically 'cold' to 'hot,' thereby promoting the recruitment and activation of dendritic cells and tumor-antigen-specific T cells. In preclinical studies, XL1350 demonstrated potent anti-tumor activity both as a single agent and in combination with immune checkpoint inhibitors. It is currently being evaluated in a Phase 1 dose-escalation clinical trial to determine its safety, tolerability, and preliminary efficacy in patients with metastatic or unresectable solid tumors.
