XL140 is a preclinical-stage PASylated protein being developed by XL-protein for the treatment of metabolic diseases. It consists of a recombinant version of insulin-like growth factor 1 (IGF-1) that has been modified using XL-protein's proprietary PASylation technology—a genetic fusion with a conformationally disordered polypeptide sequence composed of proline, alanine, and serine (PAS). This modification is designed to increase the hydrodynamic volume of the protein, thereby reducing renal clearance and significantly extending its plasma half-life compared to the native protein. XL140 acts as an IGF-1 receptor agonist, intended for use in conditions where IGF-1 replacement or supplementation is therapeutic.
