XL177B is the inactive (R)-enantiomer of XL177A, a small molecule inhibitor of the deubiquitylating enzyme USP7 (Ubiquitin-specific protease 7). Developed by researchers at the Dana-Farber Cancer Institute, XL177B is primarily utilized as a negative control in preclinical research to validate the specificity of USP7 inhibition. In experimental models of multiple myeloma, XL177B lacks the ability to induce the maturation of plasmacytoid dendritic cells (pDCs) or stimulate anti-myeloma immune responses, which are characteristic effects of its active counterpart, XL177A. Its use is critical in biochemical and cellular assays to distinguish between the targeted effects of USP7 blockade and non-specific chemical interactions.
