XLS-108 is an investigational T-cell receptor engineered T-cell (TCR-T) therapy developed by Guangdong Xiangxue Precision Medical Technology (XLifeSc). The therapy involves the genetic modification of a patient's own T cells to express a T-cell receptor (TCR) specific for a novel peptide derived from the Melanoma-associated antigen A4 (MAGE-A4), presented in the context of the HLA-A*11:01 allele. MAGE-A4 is a cancer-testis antigen that is frequently overexpressed in various solid tumors, such as esophageal cancer, non-small cell lung cancer, and head and neck squamous cell carcinoma, while its expression in normal tissues is restricted to the testes and placenta. By targeting this antigen, XLS-108 is designed to selectively recognize and eliminate MAGE-A4-positive malignant cells. As of early 2024, the candidate is in the preclinical stage of development for the treatment of multiple solid tumors.
