XmAb14702 is a bispecific monoclonal antibody developed using Xencor's XmAb bispecific Fc domain technology. It is engineered to simultaneously bind CD38, a cell surface antigen commonly expressed on multiple myeloma cells, and CD3, a component of the T-cell receptor complex. The drug is designed with low affinity for CD3 to modulate T-cell activation and cytotoxicity against CD38+ tumor cells while aiming to reduce cytokine release and improve safety compared to higher-affinity variants. Preclinical studies in primates showed that XmAb14702 had little effect on depletion of CD38+ cells or T cell activation, indicating limited potency at this affinity level[1][2][9]. The primary indication explored was multiple myeloma.
