XmAb15426 is a bispecific monoclonal antibody developed using Xencor's XmAb bispecific Fc domain technology. It is engineered to simultaneously bind CD38, a cell surface antigen highly expressed on multiple myeloma cells, and CD3, a component of the T-cell receptor complex. By engaging both targets, XmAb15426 redirects T cells to recognize and kill CD38-positive tumor cells through T cell-mediated cytotoxicity. The molecule features intermediate affinity for CD3, which in preclinical primate studies resulted in more sustained depletion of CD38+ cells compared to higher-affinity variants and was associated with lower cytokine release and improved tolerability at tested doses. The primary indication under investigation is multiple myeloma[1][2].
