Overview

XmAb18968 (also known as XmAb968 or AMG 424) is a bispecific T-cell engager antibody designed to target CD38-expressing hematologic malignancies. It simultaneously binds to CD38, a transmembrane glycoprotein highly expressed on tumor cells in conditions such as acute myeloid leukemia (AML), and CD3 on T cells, thereby redirecting T-cell cytotoxic activity to eliminate the cancer cells. The molecule features Xencor's proprietary XmAb bispecific Fc technology, which incorporates a modified Fc domain engineered to minimize binding to Fcγ receptors. This design is intended to reduce non-selective T-cell activation and cytokine release syndrome (CRS) without compromising the potent target-mediated killing of malignant cells. Originally developed by Xencor and licensed to Amgen, the rights to the program reverted to Xencor in 2020. It is currently being evaluated in Phase 1 clinical trials for relapsed or refractory AML, T-cell acute lymphoblastic leukemia (T-ALL), and T-cell lymphoblastic lymphoma (T-LBL).
