Overview

XmAb412 is a bispecific antibody being developed by Xencor for the treatment of autoimmune and inflammatory diseases, with a primary focus on inflammatory bowel disease (IBD). The therapeutic candidate is designed to simultaneously target and neutralize two key pro-inflammatory cytokines: tumor necrosis factor-like ligand 1A (TL1A) and the p19 subunit of interleukin-23 (IL-23p19). By dual-targeting these pathways, XmAb412 aims to provide enhanced efficacy compared to monotherapies targeting either pathway alone. The molecule utilizes Xencor's proprietary XmAb technology platform and incorporates an engineered Fc domain for an extended half-life, which may support less frequent dosing. As of 2025, XmAb412 is in preclinical development, with plans to initiate first-in-human clinical trials in the second half of 2026.
