Overview

XmAb541 is a first-in-class, tumor-targeted, T-cell engaging 2+1 bispecific antibody developed for the treatment of advanced solid tumors expressing Claudin 6 (CLDN6), including ovarian cancer and other CLDN6-expressing cancers. It is engineered using Xencor’s proprietary XmAb® 2+1 bispecific antibody format, which enables simultaneous binding to both the tumor-associated antigen CLDN6 and the T-cell surface antigen CD3. This dual targeting recruits and activates T cells to mediate potent and selective lysis of tumor cells expressing CLDN6 while minimizing off-tumor activity by improving selectivity over similar claudin family members such as CLDN9. The Fc domain provides extended half-life and stability. The drug is currently in Phase 1 clinical trials for multiple advanced solid tumors[1][5][6].
