XmAb564 is a monovalent interleukin-2 (IL-2) Fc fusion protein engineered by Xencor to selectively activate and expand regulatory T cells (Tregs) for the potential treatment of autoimmune diseases. It features reduced binding affinity for the IL-2 beta receptor (IL-2Rβ/CD122) and increased affinity for the alpha receptor (IL-2Rα/CD25), resulting in selective Treg expansion while minimizing activation of effector T cells and natural killer cells. The molecule incorporates a heterodimeric Fc domain with Xtend technology to provide a stable scaffold and extended half-life. Preclinical and early clinical studies have shown that XmAb564 is well tolerated, produces durable dose-dependent expansion of Tregs, and may support multi-week dosing intervals[1][3][4][5][6][8].
