XmAb662 is a potency-reduced interleukin-12 (IL-12) Fc fusion protein developed by Xencor for oncology indications. It is designed to stimulate the immune system against cancer by targeting and activating the interleukin-12 receptor (IL-12R), thereby promoting high levels of interferon gamma secretion from T cells and natural killer (NK) cells. This increases cytotoxicity and enhances tumor immunogenicity by making tumor antigens more visible to the immune system. The drug utilizes Xencor’s Xtend Fc technology to create a bispecific or heterodimeric fusion protein with improved pharmacokinetics and therapeutic potential in solid tumors[1][4][6]. Clinical development reached Phase 1 trials in advanced solid tumors but was paused as of February 2024[7].
