Drug intelligence / Profile preview
**XMAB808 + pembrolizumab** is an experimental combination therapy being studied for the treatment of advanced solid tumors. XMAB808 is a bispecific antibody developed to bind both the tumor antigen B7-H3 and the CD28 co-stimulatory receptor on T cells, providing conditional T-cell co-stimulation only at the tumor site to enhance anti-tumor immune responses while reducing off-tumor toxicity. Pembrolizumab is a monoclonal antibody that blocks the PD-1 immune checkpoint, thereby restoring T-cell activity against tumors. This combination aims to both co-stimulate tumor-infiltrating T cells (via CD28) and block PD-1-mediated immune suppression, potentially resulting in enhanced anti-tumor activity[1][2][3][4]. The combination is currently in Phase 1 clinical testing for advanced solid tumors[1][2][4].
