Overview

XmAb819 is an investigational, tumor-targeted, T-cell engaging bispecific antibody developed by Xencor for the treatment of renal cell carcinoma (RCC), specifically clear cell renal cell carcinoma (ccRCC) that is relapsed or refractory to standard therapies. It utilizes a 2+1 bispecific format with two high-avidity binding domains targeting ENPP3 (ectonucleotide pyrophosphatase/phosphodiesterase family member 3), which is highly expressed on kidney cancer cells, and one low-affinity binding domain targeting CD3 on T cells. This design enables selective engagement and activation of T cells to induce potent cytotoxicity against ENPP3-expressing tumor cells while sparing normal tissues with lower ENPP3 expression. The drug is currently being evaluated in Phase 1 clinical trials for safety, tolerability, pharmacokinetics, and preliminary efficacy in patients with advanced ccRCC[1][2][5][6][7][8].
