XmAb841 is a bispecific cytokine-Fc fusion protein developed by Xencor for the treatment of solid tumors. It is engineered to selectively deliver interleukin-15 (IL-15) to tumor-infiltrating lymphocytes (TILs) that express the checkpoint receptor Lymphocyte Activation Gene-3 (LAG-3). The molecule consists of a single-chain IL-15/IL-15Rα complex and a LAG-3-binding domain attached to a heterodimeric Fc region. By targeting LAG-3, which is frequently co-expressed with PD-1 on exhausted T cells within the tumor microenvironment, XmAb841 aims to promote the proliferation and activation of tumor-specific CD8+ T cells and natural killer (NK) cells. This targeted approach is designed to enhance anti-tumor immunity while minimizing the systemic toxicities typically associated with peripheral IL-15 activity. Additionally, the drug may exert antibody-dependent cell-mediated cytotoxicity (ADCC) against LAG-3-expressing cells.
