XMMEN-OE5 is a murine immunoglobulin M (IgM) monoclonal antibody developed to target the core lipid A region of bacterial endotoxin. It is being investigated for the treatment of suspected gram-negative bacterial sepsis. Clinical studies have shown XMMEN-OE5 to be well tolerated at doses ranging from 0.1 to 15 mg/kg when administered as a single intravenous infusion. The antibody exhibits a serum half-life of approximately 10 hours at lower doses (0.1 to 7.5 mg/kg) and about 18 hours at a higher dose (15 mg/kg). Immunogenicity, characterized by the detection of human anti-mouse antibodies, was observed in some patients receiving doses of 2.0 mg/kg or greater. Further controlled trials are indicated to establish its efficacy in treating gram-negative bacteremia.
