XMU-MP-1 is a potent, selective, and reversible small molecule inhibitor of the Hippo pathway kinases MST1 and MST2 (Mammalian Ste20-like kinases 1 and 2). By blocking the kinase activity of MST1/2, XMU-MP-1 prevents the downstream phosphorylation of LATS1/2, which in turn promotes the nuclear translocation and transcriptional activity of YAP (Yes-associated protein) and TAZ. This activation of the Hippo signaling pathway's downstream effectors facilitates tissue repair and regeneration. Originally developed by researchers at Xiamen University, XMU-MP-1 has demonstrated efficacy in promoting liver regeneration, skin wound healing, and intestinal repair in preclinical models. Recent research also indicates its potential in treating pulmonary hypertension (PH) by modulating the ARPC5/SUN1/Hippo signaling axis, thereby attenuating vascular remodeling and improving hemodynamic parameters.
