XNTR-934 is a preclinical-stage therapeutic construct being developed by Xennials Therapeutics for the treatment of prostate and breast cancers. It is designed as a multi-functional agent that combines the inhibition of the Programmed Cell Death Protein 1 (PD-1) pathway with dual inhibition of Signal Transducer and Activator of Transcription (STAT) proteins and the Androgen Receptor N-terminal domain (AR NTD). By targeting the AR NTD, XNTR-934 aims to bypass resistance mechanisms common in advanced prostate cancer, such as the expression of androgen receptor splice variants that lack the ligand-binding domain targeted by traditional therapies. The integration of a PD-1 inhibitor component is intended to stimulate an anti-tumor immune response, potentially providing a synergistic therapeutic effect when combined with the direct oncogenic pathway inhibition of STAT and AR signaling.
