XNW14010 is an orally bioavailable small molecule inhibitor specifically targeting the KRAS G12C mutation, developed by Suzhou Sinovent Pharmaceuticals. It functions by covalently binding to the cysteine residue at position 12 of the KRAS protein, effectively trapping the protein in its inactive GDP-bound state. This action prevents the activation of downstream signaling pathways, such as the MAPK/ERK pathway, which are critical for the growth and survival of cancer cells. XNW14010 is currently in Phase I/II clinical development for patients with advanced solid tumors, specifically targeting non-small cell lung cancer (NSCLC) and colorectal cancer (CRC) harboring the KRAS G12C mutation.
