XNW28012 is a next-generation antibody-drug conjugate (ADC) developed for the treatment of advanced solid tumors, including pancreatic, ovarian, and cervical cancers. The drug targets tissue factor (TF), a protein highly expressed in various malignancies. Its mechanism involves delivering a cytotoxic payload—specifically YL0014, a topoisomerase I inhibitor—directly to tumor cells expressing tissue factor. This targeted approach aims to maximize anti-tumor activity while minimizing systemic toxicity. The ADC utilizes the YiLink Bio TMALIN platform for dual-cleavage release of its payload both extracellularly in the tumor microenvironment and intracellularly within lysosomes. XNW28012 is currently being evaluated in Phase 1/2 clinical trials for safety, tolerability, pharmacokinetics, and preliminary efficacy in patients with advanced malignant solid neoplasms who have failed standard therapies or are intolerant to them[1][2][3][5][6][7].
